Application Notes
ELISA: This purified human CD120b antibody may be used in an indirect ELISA or as the capture reagent in a sandwich ELISA with a biotinylated human CD120b antibody as the detection reagent and recombinant human CD120b as the standard.
Western Blot: This purified human CD120b antibody may be used in Western Blotting applications under either reducing or non-reducing conditions with recombinant human CD120b as the positive control.
ELISA: Minimum Dilution: 0.5; Maximum Dilution: 2.0ug/ml
Functional Assays: Minimum Dilution: 15.0; Maximum Dilution: 20.0ug/ml
Immunofluorescence: Maximum Dilution: 3.0ug/ml
Western Blotting: Minimum Dilution: 0.1; Maximum Dilution: 0.2ug/ml
Reconstitution
Reconstitute with 0.1ml distilled water Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution. For long term storage the addition of 0.09% sodium azide is recommended. N.B. For functional studies do not add sodium azide

Antiserum to human CD120b were raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.

Preparation and Storage
Prior to reconstitution store at 4 degree C. After reconstitution store at -20 degree C. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.

Rabbit anti human CD120b recognizes the soluble form of human CD120b, otherwise known as sTNF receptor type II (sTNFR1I), a member of the tumour necrosis factor receptor (TNFR) superfamily, produced by proteolytic cleavage of the membrane form by the metalloprotease TACE. CD120b plays a key role in regulating the inflammatory response, acting as a receptor for both tumour necrosis factor alpha (TNF-alpha) and TNF-beta, and is also involved in programmed cell death and anti-viral activity. sTNFR1I acts as an inhibitor of TNF-alpha induced responses, by acting as a decoy receptor.

NCBI Description

The protein encoded by this gene is a member of the TNF-receptor superfamily. This protein and TNF-receptor 1 form a heterocomplex that mediates the recruitment of two anti-apoptotic proteins, c-IAP1 and c-IAP2, which possess E3 ubiquitin ligase activity. The function of IAPs in TNF-receptor signalling is unknown, however, c-IAP1 is thought to potentiate TNF-induced apoptosis by the ubiquitination and degradation of TNF-receptor-associated factor 2, which mediates anti-apoptotic signals. Knockout studies in mice also suggest a role of this protein in protecting neurons from apoptosis by stimulating antioxidative pathways. [provided by RefSeq, Jul 2008]

TNF-R2: Receptor with high affinity for TNFSF2/TNF-alpha and approximately 5-fold lower affinity for homotrimeric TNFSF1/lymphotoxin-alpha. The TRAF1/TRAF2 complex recruits the apoptotic suppressors BIRC2 and BIRC3 to TNFRSF1B/TNFR2. This receptor mediates most of the metabolic effects of TNF-alpha. Isoform 2 blocks TNF-alpha-induced apoptosis, which suggests that it regulates TNF-alpha function by antagonizing its biological activity. Binds to TRAF2. Interacts with BMX. 2 isoforms of the human protein are produced by alternative splicing.
