The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: human Jurkat cell lysates.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. (Figure. Cell proliferation of TF-1 cells after stimulated with LIF.Leukemia Inhibitory Factor (LIF), is an interleukin 6 class cytokine that affects cell growth by inhibiting differentiation. Other properties attributed to the cytokine include: the growth promotion and cell differentiation of different types of target cells, influence on bone metabolism, cachexia, neural development, embryogenesis and inflammation. p53 regulated LIF has been shown to facilitate implantation in the mouse model and possibly in humans. To test the effect of LIF on cell proliferation, TF-1 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well with 1% serum standard 1640 including various concentrations of recombinant human LIF. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of TF-1 cells after incubation with LIF for 72h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8 ) assay after incubation with recombinant LIF for 72h. The result was shown in Figure 2. (IL24 (interleukin 24) is a cytokine that belongs to IL10 family. This protein can induce apoptosis selectively in various cancer cells, including ECV304. Thus, inhibition of cell proliferation assay of IL24 was conducted using ECV-304 cells. Briefly, ECV-304 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard 1640 prior to the addition of various concentrations of IL24. (IL1alpha (Interleukin-1 alpha) is a member of the interleukin 1 cytokine family. This cytokine is produced by monocytes and macrophages as a proprotein, which is proteolytically processed and released in response to cell injury, and thus induces cell apoptosis. It is reported that exposure of MCF-7 cells to certain concentration of IL1alpha results in inhibition of cell growth. Thus, an cell proliferation assay of MCF-7 was conducted with the addition of IL1alpha. MCF-7 cells were seeded overnight at a density of 5,000 cells/well, and then treated with or without various concentrations of IL1alpha for 72h, then cells were observed by inverted microscope and cell viability was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours in at 37 degree C.Inhibition of MCF-7 cell proliferation after incubation with IL1alpha for 72h observed by inverted microscope was shown in Figure 1.)

(IL2 (Interleukin-2) is a cytokine produced by T-cells in response to antigenic or mitogenic stimulation. IL2 is a type of signaling molecule in the immune system, that is required for both T-cell and B-cell proliferation and other activities crucial to regulation of the immune response. Recombinant human IL2 shares 56% AA sequence identity with mouse IL2, suggesting the exist of cross-species activity. Therefore, in order to detect the bioactivity of rhIL2, a cell proliferation assay has been conducted using CTLL-2 mouse cytotoxic T cells. Briefly, CTLL-2 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well with or without the addition of various concentrations of IL2.
