(Interleukin-2 (IL-2), the main lymphokine produced by activated CD4 T-helper lymphocytes, is a potent T-cell growth regulator. In addition to this activity, IL-2 was found to decrease the proliferation of 3T3 fibroblasts in time- and dose-dependent manner. Thus 3T3 cells were seeded overnight at a density of 5,000 cells/well, and treated with or without various concentrations of IL2 for 48h, then 3T3 cells were observed by inverted microscope and cell viability was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37oC.The inhibitory effect of IL-2 on cell proliferation of 3T3 cells observed by inverted microscope was shown in Figure 1.)

(IL24 (interleukin 24) is a cytokine that belongs to IL10 family. This protein can induce apoptosis selectively in various cancer cells, including ECV304. Thus, inhibition of cell proliferation assay of IL24 was conducted using ECV-304 cells. Briefly, ECV-304 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard 1640 prior to the addition of various concentrations of IL24. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Inhibition of ECV-304 cells proliferation after incubation with IIL24 for 48h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8 ) assay after incubation with various concentrations of IL24 for 48h. The mean OD value of ECV-304 assessed by CCK-8 was shown in Figure 2. It was obvious that IL24 significantly decreased cell viability of ECV-304 cells.)

(IL18 (interleukin 18) is a cytokine that shares similarities in structure with IL1, and is produced by macrophages and other cells. IL18 has been reported to stimulate T cells proliferation and NK cell activity, besides, murine IL18 has been found to enchance the production of INFgamma in spleen cells in vitro. Thus, a stimulation assay was conducted to detect the activity of IL18 using murine spleen cells. Briefly, murine spleen cells were seeded into wells of 6-well plates at a density of 2×106 cells/mL in RPMI-1640 with the addition of various concentrations of IL18. After incubation for 48 hours, the concentration of INFgamma in the cell supernatant was detected using an ELISA kit. INFgamma levels in the cell supernatant of spleen cells increased significantly after stimulated with IL18, the data was shown in Figure 1.)

(Interleukin 6 (IL-6) is an interleukin that acts as both a pro-inflammatory cytokine and an anti-inflammatory myokine. Current data suggest that direct application of IL-6 on breast cancer cells inhibits proliferation in ER-positive (estrogen- receptor- positive) cells through the Jak/Stat3 pathway. (IL17RB (Interleukin-17 receptor B) is a receptor for the proinflammatory cytokines IL17B (Interleukin-17B) and IL17E (Interleukin-17E). This receptor has been shown to mediate the activation of NF-kappaB and the production of IL8 induced by IL17E. Thus a binding ELISA assay was constructed to detect the association of recombinant human IL17RB with recombinant human IL17B. Briefly, IL17RB were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL17B-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL17RB pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of IL17RB with IL17B was shown in Figure 1 and this effect was in a dose dependent manner.)

(IL1alpha (Interleukin-1 alpha) is a member of the interleukin 1 cytokine family. This cytokine is produced by monocytes and macrophages as a proprotein, which is proteolytically processed and released in response to cell injury, and thus induces cell apoptosis. It is reported that exposure of MCF-7 cells to certain concentration of IL1alpha results in inhibition of cell growth. Thus, an cell proliferation assay of MCF-7 was conducted with the addition of IL1alpha. MCF-7 cells were seeded overnight at a density of 5,000 cells/well, and then treated with or without various concentrations of IL1alpha for 72h, then cells were observed by inverted microscope and cell viability was measured by Cell Counting Kit-8 (CCK-8).
