Rat Bromodomain-containing protein 4 (BRD4) ELISA Kit
Principle of the Assay: BRD4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-BRD4 antibody and an BRD4-HRP conjugate. The assay sample and buffer are incubated together with BRD4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BRD4 concentration since BRD4 from samples and BRD4-HRP conjugate compete for the anti-BRD4 antibody binding site. Since the number of sites is limited, as more sites are occupied by BRD4 from the sample, fewer sites are left to bind BRD4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BRD4 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is homologous to the murine protein MCAP, which associates with chromosomes during mitosis, and to the human RING3 protein, a serine/threonine kinase. Each of these proteins contains two bromodomains, a conserved sequence motif which may be involved in chromatin targeting. This gene has been implicated as the chromosome 19 target of translocation t(15;19)(q13;p13.1), which defines an upper respiratory tract carcinoma in young people.
