Human Bromodomain Containing Protein 4 (BRD4) ELISA Kit
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to BRD4. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to BRD4. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain BRD4, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of BRD4 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is homologous to the murine protein MCAP, which associates with chromosomes during mitosis, and to the human RING3 protein, a serine/threonine kinase. Each of these proteins contains two bromodomains, a conserved sequence motif which may be involved in chromatin targeting. This gene has been implicated as the chromosome 19 target of translocation t(15;19)(q13;p13.1), which defines an upper respiratory tract carcinoma in young people. Two alternatively spliced transcript variants have been described. [provided by RefSeq, Jul 2008]
Uniprot Description
BRD4: an atypical protein kinase of the Brd family. Contains a double bromodomain. Binds to chromatin and regulates cell cycle progression at multiple stages. Associates with chromosomes during mitosis. The cognate gene is the chromosome 19 target of translocation t(15;19)(q13;p13.1), resulting in the BRD4-NUT oncogene identified in a lethal carcinoma of young people. Binds to papillomavirus E2 protein and is required for its transforming ability. Two alternatively spliced isoforms have been described.

Product Notes
The Human BRD4 brd4 (Catalog #AAA2088181) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2088181 ELISA Kit recognizes Human BRD4. It is sometimes possible for the material contained within the vial of "Bromodomain Containing Protein 4 (BRD4), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
