Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for ACACA elisa kit
Description: This Rat Acetyl-CoA carboxylase 1 (ACACA) ELISA Kit employs a two-site sandwich ELISA to quantitate ACACA in samples. An antibody specific for ACACA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ACACA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ACACA is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ACACA bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Acetyl-CoA carboxylase (ACC) is a biotin-dependent enzyme that catalyzes the irreversible carboxylation of acetyl-CoA to produce malonyl-CoA through its two catalytic activities, biotin carboxylase (BC) and carboxyltransferase (CT). ACC is a multi-subunit enzyme in most prokaryotes and in the chloroplasts of most plants and algae, whereas it is a large, multi-domain enzyme in the endoplasmic reticulum of most eukaryotes. The most important function of ACC is to provide the malonyl-CoA substrate for the biosynthesis of fatty acids. The activity of ACC can be controlled at the transcriptional level as well as by small molecule modulators and covalent modification. The human genome contains the genes for two different ACCs — ACACA and ACACB. In muscle cells, malonyl-CoA inhibits beta-oxidation.

NCBI Description

catalyzes the rate-limiting carboxylation of acetyl CoA to malonyl CoA during long-chain fatty acid synthesis [RGD, Feb 2006]

Uniprot Description

ACC1: a subunit of acetyl-CoA carboxylase (ACC), a multifunctional enzyme system. Catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. Phosphorylation by AMPK or PKA inhibits the enzymatic activity of ACC. ACC-alpha is the predominant isoform in liver, adipocyte and mammary gland. ACC-beta is the major isoform in skeletal muscle and heart. Phosphorylation regulates its activity.

Product Notes

The Rat ACACA acaca (Catalog #AAA9711945) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9711945 ELISA Kit recognizes Rat ACACA. It is sometimes possible for the material contained within the vial of "Acetyl-CoA Carboxylase 1 (ACACA), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
