Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-Human TMEM194A polyclonal antibody (dilution ratio 1:100) at 4 degree C overnight. Then cells were stained with the Alexa Fluor488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to Nucleus.)

Nemp1 (also known as TMEM194A), an inner nuclear membrane (INM) protein, which is expressed in the anterior neural plate in Xenopus. Nemp1 contains an evolutionarily conserved region A within its transmembrane domains (TMs) and region B within its C-terminal region, but does not contain any known domains. Nemp1 is required in oocytes of primordial follicles, which reside in an environment with high mechanical tension. Loss of high cortical tension can lead to loss of primordial follicles, and too much tension can inhibit follicle progression. NEMP1 can form a complex with Emerin and that both Emerin and NEMP1 support NE stiffness.
