Applicable Applications for anti-RPH3A antibody
Western Blot (WB)
Application Notes
Quality Control: Western blots performed on each lot.
WB: 1:1000
Antigen
Synthetic peptide corresponding to amino acid residues from the C-terminal region of rabphilin 3A.
Immunogen Information
Synthetic peptide corresponding to amino acid residues from the C-terminal region conjugated to KLH
Immunogen Species
Rat
Reactivity Assumed Based on 100% Sequence Homology
Mouse, human, bovine, canine and non-human primate
Species Reactivity Note
The antibody has been directly tested for reactivity in Western blots with rat tissue. It is anticipated that the antibody will react with mouse, human, bovine, canine and non-human primate tissue based on the fact that these species have 100% homology with the amino acid sequence used as antigen.
Biological Significance
Rabphilin-3A is a peripheral membrane protein that binds Ca2+ and phospholipids and is attached to synaptic vesicle membranes (Geppert et al., 1994;Li et al., 1994). The expression of rabphilin 3A is reduced in an animal model of Huntington's disease (Smith et al., 2005). Rabphilin can be phosphorylated at Ser234 by both CAM kinase II and PKA and this phosphorylation has been suggested to regulate neuronal activity during development in a synapse-specific manner (Fykse et al., 1995;Foletti and Scheller, 2001).
Preparation and Storage
For long term storage -20 degree C is recommended. Stable at -20 degree C for at least 1 year.

NCBI Description

binds Ca2+ and phospholipids [RGD, Feb 2006]

Uniprot Description

Function: Protein transport. Probably involved with Ras-related protein Rab-3A in synaptic vesicle traffic and/or synaptic vesicle fusion. Could play a role in neurotransmitter release by regulating membrane flow in the nerve terminal.

Subunit structure: Interacts with RAB3B, RAB3C, RAB3D, RAB8A, RAB27A and RAB27B

By similarity. Interacts with RAB3A. Monomer.

Subcellular location: Cell junction › synapse

By similarity. Membrane; Peripheral membrane protein Ref.4.

Tissue specificity: Specifically expressed in brain.

Domain: Binds calcium via the C2 domains. The calcium-bound C2 domains mediate interactions with phospholipid bilayers. Ref.4

Sequence similarities: Contains 2 C2 domains.Contains 1 FYVE-type zinc finger.Contains 1 RabBD (Rab-binding) domain.

Product Notes

The RPH3A rph3a (Catalog #AAA502108) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Rabphilin 3A reacts with Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Rabphilin 3A can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Quality Control: Western blots performed on each lot. WB: 1:1000. Researchers should empirically determine the suitability of the RPH3A rph3a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Rabphilin 3A, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
