Western Blot (WB) (Western Blot of whole rat cerebellum homogenate stained with MBS6012347 in the left strip, at dilution of 1:20,000. A prominent band running with an apparent SDS-PAGE molecular weight of ~68kD corresponds to rodent NF-L. Faint bands below this are in vivo degradation products generated by calpain digestion. The major NF-H band in human, cow and bovine run a little slower on SDS-PAGE, at about 70kD. Right lane shows a strip incubated with a new antibody against neuron specific enolase, which reveals a major band at 47kD.)

Western Blot (WB)

(Mixed neuron/glia cultures from newborn rat brain stained with 031420 antibody to peripherin (green) and rabbit polyclonal antibody to NF-L MBS6012347 (red channel). A class of large neurons, like the one in the middle of this image, contain peripherin, while the majority of neurons and their processes contain NF-L and not peripherin. Interestingly, the periperin positive cells often contain a cytoplasmic inclusion next to the nucleus which stains for both peripherin and NF-L, and so appears golden in this kind of image. The blue channel reveals the localization of DNA)

Related Product Information for anti-Neurofilament L, 68kD antibody
Neurofilaments can be defined as the intermediate or 10nm diameter filaments found in neuronal cells. They are composed a mixture of subunits which include the neurofilament triplet proteins, NF-L, NF-M and NF-H. Neurofilaments may also include other intermediate filamant subunits such as -internexin, peripherin and in developing neurons nestin and vimentin. Antibodies to neurofilament subunits are very useful cell type markers since the proteins are quite abundant, biochemically stable and found only in neurons. Mutations in the protein coding region of the human NF-L gene appear to cause some forms of Charcot-Marie-Tooth disease (2) The HGNC name for this protein is NEFL.

Product Notes

The Neurofilament L, 68kD (Catalog #AAA6012347) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Neurofilament L, 68kD (NF-L, NFL, 68kD Neurofilament Protein, NEFL, Neurofilament Light Polypeptide 68kD, NF68, Neurofilament Triplet L Protein) reacts with Porcine, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Neurofilament L, 68kD can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunocytochemistry (ICC), Immunofluorescence (IF). Suitable for use in Immunocytochemistry, Immunofluorescence and Western Blot. Dilution: Immunocytochemistry (ABC): 1:5000 Immunofluorescence: 1:500-1:1000 Western Blot: 1:20,000. Researchers should empirically determine the suitability of the Neurofilament L, 68kD for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Neurofilament L, 68kD, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
