SDS-Page (Peptide Competition Extracts prepared from NIH3T3 cells left untreated (1) or treated with PDGF (2-5) were resolved by SDS-PAGE on a 10% polyacrylamide gel and transferred to PVDF. Membranes were blocked with a 4% BSA-TBST buffer for one hour at RT then were incubated with MEK1 [pT386] antibody in a 1% BSA-TBST buffer for two hours at room temperature, following prior incubation with: no peptide (1, 2), the nonphosphopeptide corresponding to the immunogen (3), a generic phosphothreoninecontaining peptide (4), or the phosphopeptide immunogen (5). After washing, membranes were incubated with goat F(ab’)2 anti-rabbit IgG HRP conjugate and bands were detected using the Pierce SuperSignal method. The data show that only the peptide corresponding to MEK1 [pT386] blocks the antibody signal, thereby demonstrating the specificity of the antibody.)

Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Member of MAP Kinase Kinase. Likely functions in a stress-activated MAPK pathway. Can phosphorylate the MAPK AtMPK4, in response to stress. Gets phosphorylated by MEKK1 in response to wounding.

Function: May be involved in the cold and salinity stress-mediated MAP kinase signaling cascade (MEKK1, MEK1/MKK2 and MPK4/MPK6). Phosphorylates MPK4 in vitro. Ref.8

Catalytic activity: ATP + a protein = ADP + a phosphoprotein.

Enzyme regulation: Activated through serine and threonine phosphorylation in response to wounding, cold, drought, salt stresses, hydrogen peroxide, bacterial flagellin and laminarin beta-glucan. Ref.10 Ref.12

Subunit structure: Interacts with MEKK1 (MAPKKK) and MPK4. May form a ternary complex with. Ref.7
