Preparation and Storage
This product is stable for several weeks at 4 degree C as an undiluted liquid. Dilute only prior to immediate use.

Description: The antibody is a rabbit polyclonal antibody made to the serine proteinase kallikrein-10 (hK10). The antibody is made to a synthetic peptide based on the Kallikrein loop area of human Kallikrein-10. The antibody reacts with human Kallikrein-10, and does not react with the human kallikreins (hK1-15). The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant. Kallikrein-10, also known as normal epithelial cell specific-1 (NES1) and protease serine-like 1 (PRSSL1), is a trypsin-like serine proteinase. Originally described from a subtractive library looking for genes that were down-regulated in breast cancer, kallikrein-10 was thought to be epithelial cell specific. Kallikrein-10 is reported to be up regulated in pancreatic cancer and ovarian cancer, but down regulated in testicular cancer and breast cancer, perhaps due to differential hormonal regulation, or tissue specificities. Breast cancer patients that had elevated kallikrein-10 had a worse prognosis, leading to speculation that kallikrein-10 might be protective in breast cancer, similar to the argument put forth for kallikrein-3 in prostate cancer. Kallikrein-10 is found in high levels in the pancreas, and in lower levels in the tonsils, esophagus, skin, brain, testis and ovary. Biological fluids (breast milk, serum, seminal plasma, urine) contain kallikrein-10, the highest levels found in milk. The cerebral spinal fluid (CSF) contains Kallikrein-10, and was reported to be elevated in patients with Alzheimer's disease. Endogenous inhibitors include kallistatin, protein-C inhibitor and beta1-proteinase inhibitor, although Kallikrein-10 can be found complexed to a number of different proteinase inhibitors. Pre-pro-kallikrein-10 has the 33 amino acid signal sequence is removed before secretion, and the Pro-kallikrein-10 is activated to Kallikrein-10 by removal of the 10 amino acid propeptide domain. The full length Kallikrein-10 sequence codes for a protein of 276 amino acids, with predicted mass of 30.1kDa, and a pI of 9.78. A range of different sized bands can be seen for Kallikrein-10 in different tissues and in cell culture media, ranging from 16-34 kDa, and likely representing different proteolytically processed forms. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species.
