Background: Glycogen synthase kinase 3-alpha is a multifunctional protein serine kinase. GSK-3 phosphorylates and inactivates glycogen synthase. It has also been implicated in the regulation of cell fate in Dictyostelium and is a component of the Wnt signaling pathway required for Drosophila, Xenopus, and mammalian development. GSK-3 has been shown to regulate Cyclin D1 proteolysis and subcellular localization. GSK-3alpha (GSK-3alpha) regulates the production of amyloid-beta peptides, a major component of the plaques that accumulate with progression of Alzheimer's disease, by phosphorylating Tau proteins. This presents the possibility that defects in GSK-3 signaling can contribute to the onset of Alzheimer's disease. GSK-3alpha can be phosphorylated by Akt at Ser21. This phosphorylation of GSK-3alpha negatively regulates its kinase activity.

GSK3A: a proline-directed protein kinase of the GSK family. Implicated in the control of several regulatory proteins including glycogen synthase, Myb, and c-Jun. GSK3 and GSK3 have similar functions. GSK3 phophorylates tau, the principal component of neurofibrillary tangles in Alzheimer disease and is required for maximal production of amyloid plaque peptides by secretase. A GSK3 promoter SNP effects progression of bipolar disorder. The GSK3 inhibitor, lithium, is used to treat bipolar disorder and is seen to block plaque formation. GSK3 generally opposes the action of insulin, and GSK3 hyperactivity is thought to contribute to insulin resistant (type II) diabetes. GSK3 also negatively regulates cardiac hypertrophy.
