Synthetic phosphopeptide (KLH coupled) corresponding to residues surrounding Ser21 of human GSK-3alpha.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Glycogen synthase kinase 3 (GSK-3) was initially identified as an enzyme that regulates glycogen synthesis in response to insulin (1). GSK-3 is a ubiquitously expressed serine/threonine protein kinase that phosphorylates and inactivates glycogen synthase. GSK-3 is a critical downstream element of the PI3 kinase/Akt cell survival pathway, and its activity can be inhibited by Akt-mediated phosphorylation at Ser21 of GSK-3alpha and Ser9 of GSK-3beta (2,3). GSK-3 has been implicated in the regulation of cell fate in Dictyostelium and is a component of the Wnt signaling pathway required for Drosophila, Xenopus and mammalian development (4). GSK-3 has been shown to regulate cyclin D1 proteolysis and subcellular localization (5).

Glycogen synthase kinase 3-alpha (GSK3A; EC {2.7.1.37}) is a multifunctional protein serine kinase, homologous to Drosophila 'shaggy' (zeste-white3) and implicated in the control of several regulatory proteins including glycogen synthase (see GYS1, {138570}) and transcription factors (e.g., JUN, {165160}). It also plays a role in the WNT ({164820}) and PI3K (see PIK3CG, {601232}) signaling pathways (see review by {1:Ali et al. (2001)}).[supplied by OMIM]

Uniprot Description

GSK3A: a proline-directed protein kinase of the GSK family. Implicated in the control of several regulatory proteins including glycogen synthase, Myb, and c-Jun. GSK3 and GSK3 have similar functions. GSK3 phophorylates tau, the principal component of neurofibrillary tangles in Alzheimer disease and is required for maximal production of amyloid plaque peptides by secretase. A GSK3 promoter SNP effects progression of bipolar disorder. The GSK3 inhibitor, lithium, is used to treat bipolar disorder and is seen to block plaque formation. GSK3 generally opposes the action of insulin, and GSK3 hyperactivity is thought to contribute to insulin resistant (type II) diabetes. GSK3 also negatively regulates cardiac hypertrophy. A tumor suppressor role is indicated by the oncogenic potential of stabilized -catenin mutants that lack GSK3 phosphorylation sites. Inhibitor: AR-A014418
