Application Notes
ELISA: This purified human EGF antibody may be used in an indirect ELISA or as the capture reagent in a sandwich ELISA with a biotinylated human EGF antibody as the detection reagent and recombinant human EGF as the standard.
Western Blot: Rabbit anti human EGF may be used in Western Blotting applications under either reducing or non-reducing conditions with recombinant human EGF as the positive control.
ELISA: Minimum Dilution: 0.5; Maximum Dilution: 2.0ug/ml
Functional Assays: Maximum Dilution: <0.1ug/ml
Immunohistology - Paraffin: Maximum Dilution: 5.0ug/ml; Application Note: This product requires heat-mediated pre-treatment of paraffin sections prior to staining. Citrate buffer pH 6.0 is recommended for this purpose
Western Blotting: Minimum Dilution: 0.1; Maximum Dilution: 0.2ug/ml
Reconstitution
Reconstitute with 0.1ml distilled water.For long term storage the addition of 0.09% sodium azide is recommended.N.B. For functional studies do not add azide.Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution.

Preparation and Storage
Prior to reconstitution store at 4 degree C. After reconstitution store at -20 degree C. Storage in frost-free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.

Rabbit anti human EGF recognizes human Epidermal Growth Factor, a potent stimulator and regulator of the proliferation of epidermal and epithelial cells, which is expressed by a variety of cells.. The effects of EGF are initiated through tyrosine kinase activity, following the binding of EGF to the extracellular domain of the 170kDa EGF receptor (EGFR), which also binds other EGF-like ligands with high affinity, including TGF-a, VGF (vaccinia virus growth factor) and Betacellulin, produced by certain tumour cells. Receptor activation through EGF binding triggers several signal transduction pathways, including the JAK/STAT and P13K/AKT pathways.

NCBI Description

This gene encodes a member of the epidermal growth factor superfamily. The encoded protein is synthesized as a large precursor molecule that is proteolytically cleaved to generate the 53-amino acid epidermal growth factor peptide. This protein acts a potent mitogenic factor that plays an important role in the growth, proliferation and differentiation of numerous cell types. This protein acts by binding the high affinity cell surface receptor, epidermal growth factor receptor. Defects in this gene are the cause of hypomagnesemia type 4. Dysregulation of this gene has been associated with the growth and progression of certain cancers. Alternate splicing results in multiple transcript variants.[provided by RefSeq, May 2010]

EGF: EGF stimulates the growth of various epidermal and epithelial tissues in vivo and in vitro and of some fibroblasts in cell culture. Magnesiotropic hormone that stimulates magnesium reabsorption in the renal distal convoluted tubule via engagement of EGFR and activation of the magnesium channel TRPM6. Defects in EGF are the cause of hypomagnesemia type 4 (HOMG4); also known as renal hypomagnesemia normocalciuric. HOMG4 is a disorder characterized by massive renal hypomagnesemia and normal levels of serum calcium and calcium excretion. Clinical features include seizures, mild-to mederate psychomotor retardation, and brisk tendon reflexes. 2 isoforms of the human protein are produced by alternative splicing.
