Application Notes
ELISA: This purified human IL-17 antibody may be used in an indirect ELISA or as the capture reagent in a sandwich ELISA with our biotinylated human IL-17 antibody as the detection reagent and recombinant human IL-17 as the standard.
Western Blot: This antibody may be used in Western Blotting under either reducing or non-reducing conditions with recombinant human IL-17 as the positive control.
ELISA: Minimum Dilution: 0.5; Maximum Dilution: 2.0ug/ml
Functional Assays: Minimum Dilution: 0.93; Maximum Dilution: 1.3ug/ml; Application Note: To yield one-half maximal inhibition [ND50] of the biological activity of hIL-17A (50.0 ng/ml).
Immunohistology - Paraffin: Maximum Dilution: 0.25ug/ml; Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose.
Western Blotting: Minimum Dilution: 0.1; Maximum Dilution: 0.2ug/ml
Reconstitution
Reconstitute with 0.1ml distilled water Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution. For long term storage the addition of 0.09% sodium azide is recommended. N.B. For functional studies do not add sodium azide

Preparation and Storage
Prior to reconstitution store at 4 degree C. Following reconstitution store at -20 degree C. This product should be stored undiluted. Storage in frost-free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.

Rabbit anti Human Interleukin-17 antibody recognizes human IL-17 (Interleukin-17/17A), a 32kDa disulfide-linked homodimer consisting of two 136 amino acid chains, which belongs to the IL-17 family (classified as IL-17A to IL-17F), and signals through the IL-17 receptor (IL-17R/CDw217). IL-17 is produced primarily by activated CD4+ T cells and induces a variety of target cells to secrete inflammatory cytokines, including IL-1beta, IL-6, TNFalpha, IFNgamma and granulocyte colony-stimulating factor, as well as synergizing with other inflammatory cytokines and agonists. The role of IL-17 in bone metabolism has become a focus area for studies into Rheumatoid Arthritis, in which levels of IL-17 are significantly increased and may contribute to bone destruction.

NCBI Description

The protein encoded by this gene is a proinflammatory cytokine produced by activated T cells. This cytokine regulates the activities of NF-kappaB and mitogen-activated protein kinases. This cytokine can stimulate the expression of IL6 and cyclooxygenase-2 (PTGS2/COX-2), as well as enhance the production of nitric oxide (NO). High levels of this cytokine are associated with several chronic inflammatory diseases including rheumatoid arthritis, psoriasis and multiple sclerosis. [provided by RefSeq, Jul 2008]
