Application Notes
Flow Cytometry: 2.5ug/1x10e6 cells using MCF-7 human breast cancer cell line treated with pervanadate, fixed with paraformaldehyde and permealized with methanol.

Immunohistochemistry: 5-15ug/ml using immersion fixed, frozen section of mouse embryo.

Western Blot: 0.5ug/ml using pervanadate-treated MDA-MB-468 human breast cancer cell line .

Optimal dilutions should be determined by each laboratory for each application.

Preparation and Storage
Lyophilized powder may be stored at -20°C. Reconstitute with sterile buffer. Aliquot and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Immunohistochemistry (IHC)

(Immunhistochemical staining of immersion fixed, frozen sections of mouse embryo (13 d.p.c) using MBS629762 at 15ug/ml (overnight incubation at 4°C. Tissue was stained using an anti-rabbit HRP-DAB staining kit and counterstained with hematoxylin (blue).)

Western Blot (WB)

(Western blot shows immunoprecipitate of MDA-MB-468 human breast cancer cell line untreated (-) or treated (+) with 100uM pervanadate for 10 mins. PVDF membrane was probed with 0.5ug/ml MBS629762 , followed by HRP-conjugated anti-rabbit IgG secondary antibody. A specific band was detected for phospho HGF R/cMet (Y1234/Y1235) at ~145kD. Experiment was conducted under reducing conditions.)

Hepatocyte growth factor receptor (HGF R), also known as c-MET, is a receptor tyrosine kinase that consists of an extracellular chain and a longer chain. The chain contains the transmembrane helix and the cytoplasmic portion. Ligand-stimulated phosphorylation of tyrosine 1234 and tyrosine 1235 in HGF R's chain results in stimulation of intrinsic kinase activity.

Uniprot Description

Function: Receptor for hepatocyte growth factor and scatter factor. Has a tyrosine-protein kinase activity. Functions in cell proliferation, scattering, morphogenesis and survival

Catalytic activity: ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate.

Subunit structure: Heterodimer formed of an alpha chain (50 kDa) and a beta chain (145 kDa) which are disulfide linked. Binds PLXNB1 and GRB2. Interacts with INPP5D/SHIP1. When phosphorylated at Tyr-1357, interacts with INPPL1/SHIP2. Interacts with RANBP9 and RANBP10, as well as SPSB1, SPSB2, SPSB3 and SPSB4. SPSB1 binding occurs in the presence and in the absence of HGF, however HGF treatment has a positive effect on this interaction. Interacts with MUC20; prevents interaction with GRB2 and suppresses hepatocyte growth factor-induced cell proliferation. Interacts with GRB10

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "c-Met, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage.
