Principle of the assay: PLAP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PLAP antibody and an PLAP-HRP conjugate. The assay sample and buffer are incubated together with PLAP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PLAP concentration since PLAP from samples and PLAP-HRP conjugate compete for the anti-PLAP antibody binding site. Since the number of sites is limited, as more sites are occupied by PLAP from the sample, fewer sites are left to bind PLAP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PLAP concentration in each sample is interpolated from this standard curve.

The first three are located together on chromosome 2 while the tissue non-specific form is located on chromosome 1. The product of this gene is a membrane bound glycosylated enzyme, also referred to as the heat stable form, that is expressed primarily in the placenta although it is closely related to the intestinal form of the enzyme as well as to the placental-like form. The coding sequence for this form of alkaline phosphatase is unique in that the 3' untranslated region contains multiple copies of an Alu family repeat.
