Principle of the assay: PDGFD ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for PDGFD. Standards or samples are then added to the microtiter plate wells and PDGFD if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of PDGFD present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for PDGFD are added to each well to “sandwich” the PDGFD immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain PDGFD and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PDGFD concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a core motif of eight cysteines, seven of which are found in this factor. This gene product only forms homodimers and, therefore, does not dimerize with the other three family members. It differs from alpha and beta members of this family in having an unusual N-terminal domain, the CUB domain. Two splice variants have been identified for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

PDGFD: Growth factor that plays an essential role in the regulation of embryonic development, cell proliferation, cell migration, survival and chemotaxis. Potent mitogen for cells of mesenchymal origin. Plays an important role in wound healing. Induces macrophage recruitment, increased interstitial pressure, and blood vessel maturation during angiogenesis. Can initiate events that lead to a mesangial proliferative glomerulonephritis, including influx of monocytes and macrophages and production of extracellular matrix. Belongs to the PDGF/VEGF growth factor family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Oncoprotein; Secreted, signal peptide; Secreted

Chromosomal Location of Human Ortholog: 11q22.3

Cellular Component: Golgi membrane; endoplasmic reticulum lumen; extracellular region

Molecular Function: growth factor activity; platelet-derived growth factor receptor binding

Biological Process: positive regulation of cell division; multicellular organismal development; platelet-derived growth factor receptor signaling pathway; regulation of peptidyl-tyrosine phosphorylation

Product Notes

The Monkey PDGFD pdgfd (Catalog #AAA737062) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA737062 ELISA Kit recognizes Monkey PDGFD. It is sometimes possible for the material contained within the vial of "Platelet Derived Growth Factor D, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
