Short Description: Recombinant METTL11A/NTMT1 protein was expressed in a baculovirus expression system as the full-length protein (accession number NP_054783.2) with an N-terminal DYKDDDDK tag. The molecular weight of the protein is 27 kDa. It is suitable for use in in binding assays, inhibitor screening, and selectivity profiling.

Background: METTL11A (Methyltransferase-Like Protein 11A)/NTMT1 (N-Terminal Xaa-Pro-Lys N-Methyltransferase 1), also known as NRMT1 or NTM1A, is a distributive alpha-N-methyltransferase that methylates the N-terminus of target proteins containing the N-terminal motif [Ala/Gly/Pro/Ser]-Pro-Lys when the initiator Met is cleaved. It specifically catalyzes mono-, di- or tri-methylation of the exposed alpha-amino group of the Ala, Gly or Ser residue in the [Ala/Gly/Ser]-Pro-Lys motif and mono- or di-methylation of Pro in the Pro-Pro-Lys motif. Some of the substrates may be primed by METTL11B-mediated monomethylation. METTL11A catalyzes the trimethylation of the N-terminal Gly in CENPA (after removal of Met- 1). It is also responsible for the N-terminal methylation of KLHL31, MYL2, MYL3, RB1, RCC1, RPL23A and SET. And it is required during mitosis for normal bipolar spindle formation and chromosome segregation via its action on RCC1.

NCBI Description

The METTL11A gene encodes an N-terminal methyltransferase for the RAN (MIM 601179) guanine nucleotide exchange factor regulator of chromosome condensation 1 (RCC1; MIM 179710). METTL11A enzyme alpha-N-methylates other protein targets such as SET (MIM 600960) and RB (MIM 180200).[supplied by OMIM, Nov 2010]

Uniprot Description

METTL11A: Alpha-N-methyltransferase that methylates the N-terminus of target proteins containing the N-terminal motif [Ala/Pro/Ser]- Pro-Lys when the initiator Met is cleaved. Specifically catalyzes mono-, di- or tri-methylation of exposed alpha-amino group of Ala or Ser residue in the [Ala/Ser]-Pro-Lys motif and mono- or di- methylation of Pro in the Pro-Pro-Lys motif. Responsible for the N-terminal methylation of KLHL31, MYL2, MYL3, RB1, RCC1, RPL23A and SET. Required during mitosis for normal bipolar spindle formation and chromosome segregation via its action on RCC1. Belongs to the methyltransferase superfamily. NTM1 family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The METTL11A ntmt1 (Catalog #AAA389088) is a Recombinant Protein produced from Baculovirus and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "METTL11A, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
