Preparation and Storage
For unopened kit, all reagents should be kept according to the labels on vials. The TMB Substrate, Wash Buffer, Stop Solution should be stored at 4 degree C. All others should be stored at -20 degree C.

Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of APLN in mouse serum, plasma and other biological fluids.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to APLN. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to APLN. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain APLN, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of APLN in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

This gene encodes the neuropeptide Apelin, an endogenous ligand of the G protein-coupled receptor APJ. This gene is highly expressed in the central nervous system and peripheral tissues and the encoded preproprotein undergoes proteolytic processing to generate mature peptides of different sizes that are secreted into the plasma. These peptides play important roles in a broad range of physiological processes such as cardiac contractility, blood pressure, blood vessel growth, appetite and drink behavior, and pituitary hormone secretion. Mice lacking the encoded protein develop progressive heart failure and exhibit a shorter bleeding time, prothrombotic profile and increased bone mass. [provided by RefSeq, Jul 2016]

Uniprot Description

Endogenous ligand for APJ. Has an inhibitory effect on cytokine production in response to T-cell receptor/CD3 cross-linking. The oral intake of apelin in the colostrum and the milk might therefore modulate immune responses in neonates. May also have a role in the central control of body fluid homeostasis by influencing AVP release and drinking behavior.
