Immunofluorescence (IF) (Immunofluorescence staining of mouse splenocytes using anti-TWEAK antibody MTW-1. Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes immobilized on Shi-fix cover-slips and stained with the chimeric rabbit IgG version of MTW-1 at 10ug/ml followed by Alexa Fluor 488 secondary antibody (2ug/ml), showing membrane staining in subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom, DAPI, merged channels and an isotype control. The isotype control was stained with anti-Fluorescein antibody followed by Alexa Fluor 488 secondary antibody.)

Application Notes
The specificity of this antibody has been confirmed in flow cytometric analysis, where it reacts specifically with mTWEAK-transfected L5178Y cells, and not with parental L5178Y cells or cells transfected with related TNF family members (Nakayama et al, 2003). Additionally, this antibody has been used to assess TWEAK cell surface expression in flow cytometric analysis (Nakayama et al, 2003). This antibody has been shown to block CD8-mTWEAK cytotoxicity against L5178Y cells over-expressing murine Fn14, the TWEAK receptor (Nakayama et al, 2003). When administered to collagen-induced arthritis (CIA) mice, this blocking antibody has been shown to have anti-inflammatory and antiangiogenic effects, with associated improvements to both the clinical and histological manifestations of CIA (Kamata et al, 2006). Additionally, administration of this antibody to denervated mice attenuates denervation-induced skeletal muscle loss (Mittal et al, 2010). Finally, administration of this antibody to CD163-/- mice subject to hind limb ischaemia inhibits the increased perfusion resulting from CD163 deficiency (Akahori et al, 2015).
Antibody Type
Recombinant Antibody
Immunogen
This antibody was raised by immunising SD rats with CD8-mTWEAK, a fusion of the human CD8alpha extracellular region to murine TWEAK.
Chimeric Use Statement
This chimeric rabbit antibody was made using the variable domain sequences of the original Rat IgG1 format, for improved compatibility with existing reagents, assays and techniques.
Preparation and Storage
Store at 4 degree C for up to 3 months. For longer storage, aliquot and store at -20 degree C.

Product Notes

The TWEAK tnfsf12 (Catalog #AAA488532) is an Antibody produced from Rat and is intended for research purposes only. The product is available for immediate purchase. The Anti-TWEAK [MTW-1], Rabbit IgG, Kappa reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's TWEAK can be used in a range of immunoassay formats including, but not limited to, Blocking (BL), Flow Cytometry (FC/FACS). The specificity of this antibody has been confirmed in flow cytometric analysis, where it reacts specifically with mTWEAK-transfected L5178Y cells, and not with parental L5178Y cells or cells transfected with related TNF family members (Nakayama et al, 2003). Researchers should empirically determine the suitability of the TWEAK tnfsf12 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TWEAK, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
