Rat anti-Mouse Tim-3 Monoclonal Antibody | anti-Tim-3 antibody
Anti-Tim-3 [2C12]
Immunofluorescence (IF)
(Immunofluorescence staining of fixed RAW264.7 cells with anti-Tim-3 antibody 2C12 (MBS488620) Immunofluorescence analysis of paraformaldehyde fixed RAW264.7 cells on Shi-fix coverslips stained with the chimeric rabbit IgG version of 2C12 (MBS488620) at 10ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom MBS488620, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (MBS488140) followed by staining with Alexa Fluor 488 secondary antibody.)
This antibody was raised by immunising rats with TIM-3:Ig fusion protein emulsified in complete Freund's adjuvant.
Note on publication: Describe the generation of this antibody and its subsequent use in FACS, immunohistochemistry, immunofluorescence and a murine experimental autoimmune encephalomyelitis (EAE) model to study the effects of TIM-3 triggering in dendritic cells.

Product Notes
The Tim-3 havcr2 (Catalog #AAA488621) is an Antibody produced from Rat and is intended for research purposes only. The product is available for immediate purchase. The Anti-Tim-3 [2C12] reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Tim-3 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunofluorescence (IF), Immunohistochemistry (IHC), Blocking (BL), Therapeutics. This antibody has been used in multiple FACS analyses for diverse immuno-oncological applications, such as to delineate how TIM-3 signaling on cells of the innate immune system critically influences regulation of the adaptive immune response (Frisancho-Kiss et al, 2006), to demonstrate the reversal of T cell exhaustion and restoration of anti-tumor immunity upon co-targeting Tim-3 and PD-1 pathways (Sakuishi et al, 2010), and to examine the therapeutic potential of dendritic cell based vaccines against malignant glioma (Dey et al, 2015). This antibody has also been used in immunohistochemistry to aid the elucidation of the effects of Th2 immune deviation on corneal allograft survival and possible mechanisms of graft rejection (Beauregard et al, 2005). In addition, employing this anti-TIM-3 antibody as part of the combination immuntherapy (anti-TIM3 + anti-PD-1 +/- anti-CTLA-4) has been shown to be effective against experimental and carcinogen-induced tumors in mice (Sakuishi et al, 2010; Ngiow et al, 2011). Researchers should empirically determine the suitability of the Tim-3 havcr2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Tim-3, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
