Specificity
Recognizes phosphorylated Ser or Thr that is surrounded by Tyr or Phe at the -1 and +1 positions and Phe at the -4 position. It also recognizes peptides containing Lys instead of Phe at the -4 position. Does not crossreact with the non-phosphorylated PDK1 docking motifs or with other phosphorylated motifs. This antibody detects endogenous levels of phosphorylated proteins containing PDK1 docking motif, including phospho-Akt.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

A hallmark of signal transduction pathways is the reversible phosphorylation of serine and threonine residues within specific sequences, or motifs, in target proteins. Specific signaling motifs include not only sequences that are recognized by protein kinases (1), but also those that are recognized by phosphorylation-dependent binding proteins like 14-3-3 (2). These modular phosphoprotein interacting domains are critical elements in modulating, directing and amplifying intracellular communications. Our phospho-motif specific antibodies provide new tools for probing the complexity of phosphoregulatory pathways. Many critical protein kinases can be regulated by phosphorylation at a specific serine or threonine in a hydrophobic motif (3). For example, Akt, an important kinase that regulates cell survival, is activated by phosphorylation at Ser473, a site preceded by phenylalanine at -4 and -1 and followed by tyrosine at +1 (4). RSK2, p70 S6 K and certain PKC isoforms also contain a similar consensus phosphorylation motif. Phosphorylation of these motifs is required for binding to 3-phosphoinositide-dependent kinase 1 (PDK1) (5-7). This antibody is a powerful tool for the characterization of phosphorylated PDK1 docking motifs and the identification of new proteins with PDK1 docking motifs.
