• Specificity: Recognizes acetylated lysine-containing proteins including histones. Of all the possible H3 and H4 acetylation sites, this antibody exhibits highest affinity for histone H4 acetylated on lysine 8 and histone H3 acetylated on lysine 14.

• Application Notes: Suitable for use in Western Blot and Immunoprecipitation.; Dilution: Western Blot: 0.5-2ug/ml detects acetylated histones in acid-extracted proteins from sodium butyrate (5mM) treated HeLa cells. Sodium butyrate, an inhibitor of deacetylases, was used to increase the level of acetylated histones. 0.5-4ug/ml detected: chemically acetylated BSA (10ng), GST-p53 (400ng) acetylated in vitro with either recombinant p300 or PCAF, auto-acetylated p300 (400ng) and PCAF (400ng). Proteins were resolved by electrophoresis, transferred to nitrocellulose and probed with 4ug/ml L9058-02A. Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.; Immunoprecipitation: 5ug immunoprecipitates in vitro acetylated PCAF added to a 3T3 RIPA cell lysate. The immunoprecipitated PCAF was detected by subsequent immunoblot analysis using 1ug/ml monoclonal anti-GST.

• Preparation and Storage: May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
