Immunofluorescence (IF) (Immunofluorescence staining of MCF7 cells using anti-c-erbB-2 FWP51 Immunofluorescence analysis of paraformaldehyde fixed MCF7 cells stained with the chimeric mouse IgG version of FWP51 at 10ug/ml followed by Alexa Fluor 488 secondary antibody (2ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom, DAPI, merged channels and an isotype control. The isotype control was stained with anti-unknown antibody followed by Alexa Fluor 488 secondary antibody.)

Application Notes
Immunofluorescence and immunoprecipitation were used to confirm the specificity of FWP51 for c-erbB-2 (Harwerth et al, 1992). Flow cytometry showed that FWP51 binds the extracellular domain of c-erbB-2 (Harwerth et al, 1992). FWP51 has also been fused to the bacterial toxin, exotoxin A. The resultant fusion protein Fv(FWP51)-ETA selectively inhibited protein synthesis in cells expressing high levels of human c-erbB-2 protein, but not in cells expressing little to no human c-erbB-2 protein. Administration of FWP51 to mice impanted with human erbB-2 transformed NIH3T3 cells led to inhibition of the onset of tumour growth (Harwerth et al, 1993).
Antibody Type
Recombinant Antibody
Immunogen
20x10^6 SKBR3 human breast tumour cells were injected into BALB/c mice. The hybridomas generated from the mice spleen cells were then screen for antibodies reactive to c-erbB-2 using immunofluorescence and immunoprecipitation.
Preparation and Storage
Store at 4 degree C for up to 3 months. For longer storage, aliquot and store at -20 degree C.

Product Notes

The c-erbB-2 (Catalog #AAA488526) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Anti-c-erbB-2 [FWP51], Mouse IgG1, Kappa reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's c-erbB-2 can be used in a range of immunoassay formats including, but not limited to, Fusion, Immunoprecipitation (IP), Flow Cytometry (FC/FACS), Immunofluorescence (IF). Researchers should empirically determine the suitability of the c-erbB-2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "c-erbB-2, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
