Host
Mouse
Reactivity
Human
Clonality
Monoclonal
Isotype
Mouse IgG1, Kappa
Clone Number
AHT107
Specificity
AHT107 binds to a different epitope than IL-2. Suggested that determinant recognised by AHT107 is involved in formation of the high-affinity binding site of the p55 + p75 heterodimeric complex of the IL-2R in a way which does not disturb the interaction of the p55 molecule and IL-2, and that the high-affinity class of the IL-2R is responsible for mediating the growth signal at physiological concentrations of IL-2.
Purity/Purification
Protein A affinity purified
Form/Format
PBS with 0.02% Proclin 300
Concentration
1mg/ml (varies by lot)
Applicable Applications for anti-IL2R antibody
Blocking (BL), ELISA (EIA), Flow Cytometry (FC/FACS)
Application Notes
This antibody has been used to create a chimeric antibody with a murine variable region and human constant region through the use of recombinant DNA techniques. This gave antigen specificity of the mouse, and immunogenicity, serum half-life, and effector functions of human antibodies. The chimeric antibody has been found to have a greater anti-proliferative effect on T lymphocytes (Rose 1991). AHT107 has been used in ELISAs for the detection of soluble human IL-2R in response to different forms of helminthic and protozoal infections (O. Josimovic et al. 1988).
Antibody Type
Recombinant Antibody
Immunogen
BALB/c mice were immunized with human T lymphoblasts and spleen cells of the immunized animals were fused with X63-Ag8.653 mouse myeloma cells. The supernatants of the resultant hybridomas were screened by a proliferation inhibition assay (inhibition of the human T lymphoblast response to IL-2). Two of those (the clones AHT54 and AHT107) that produced antibodies which inhibited IL-2 dependent proliferation of human T lymphoblasts were selected, cloned and recloned.
Preparation and Storage
Store at 4 degree C for up to 3 months. For longer storage, aliquot and store at -20 degree C.

NCBI Description

The interleukin 2 (IL2) receptor alpha (IL2RA) and beta (IL2RB) chains, together with the common gamma chain (IL2RG), constitute the high-affinity IL2 receptor. Homodimeric alpha chains (IL2RA) result in low-affinity receptor, while homodimeric beta (IL2RB) chains produce a medium-affinity receptor. Normally an integral-membrane protein, soluble IL2RA has been isolated and determined to result from extracellular proteolyisis. Alternately-spliced IL2RA mRNAs have been isolated, but the significance of each is presently unknown. Mutations in this gene are associated with interleukin 2 receptor alpha deficiency.[provided by RefSeq, Nov 2009]

Uniprot Description

IL2RA: Receptor for interleukin-2. Non-covalent dimer of an alpha and a beta subunit. IL2R exists in 3 different forms: a high affinity dimer, an intermediate affinity monomer (beta subunit), and a low affinity monomer (alpha subunit). The high and intermediate affinity forms also associate with a gamma subunit.

Product Notes

The IL2R il2ra (Catalog #AAA488502) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Anti-IL2R [AHT107], Mouse IgG1, Kappa reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's IL2R can be used in a range of immunoassay formats including, but not limited to, Blocking (BL), ELISA (EIA), Flow Cytometry (FC/FACS). 1988). Researchers should empirically determine the suitability of the IL2R il2ra for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IL2R, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
