A66 (p110alpha inhibitor A66) is a potent, highly specific p110alpha inhibitor with IC50 of 32, 30 and 43nM for wt p110alpha, p110alpha E545K and p110alpha H1047R, respectively; displays little to no activity against p110beta/gamma/delta (IC50>3,000nM), as well as class-III PI3Ks, mTOR, DNA-PK, and PI4Kalpha/beta; induces reduction in focus formation CEF by the highly transforming iSH2 mutants KS459delN, DKRMNS560del, and K379E, blocks PI3K signalling and tumour growth in certain cell types.
