Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Histone H2B type 2-E. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Histone H2B type 2-E and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Histone H2B type 2-E, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Histone H2B type 2-E in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene encodes a replication-dependent histone that is a member of the histone H2B family, and generates two transcripts through the use of a putative stem-loop termination motif, and the polyA addition motif. Uniprot Description

H2B1L: a core component of the nucleoosome. The nucleosome, a basic organizational unit of chromosomal DNA, is octrameric, consisting of two molecules each of histones H2B, H2A, H3, H4. The octamer wraps approximately 147 bp of DNA. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.

Product Notes

The Mouse Hist2h2be hist2h2be (Catalog #AAA288329) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA288329 ELISA Kit recognizes Mouse Hist2h2be. It is sometimes possible for the material contained within the vial of "Histone H2B type 2-E, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
