Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Neuropeptide FF receptor 2. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Neuropeptide FF receptor 2 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Neuropeptide FF receptor 2, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Neuropeptide FF receptor 2 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Uniprot Description

NPFFR2: Receptor for NPAF (A-18-F-amide) and NPFF (F-8-F-amide) neuropeptides, also known as morphine-modulating peptides. Can also be activated by a variety of naturally occurring or synthetic FMRF-amide like ligands. This receptor mediates its action by association with G proteins that activate a phosphatidylinositol- calcium second messenger system. Belongs to the G-protein coupled receptor 1 family. 4 isoforms of the human protein are produced by alternative splicing.
