Principle of the Assay: The ELISA is based on the competitive binding enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Histone acetyltransferase KAT8, During the reaction, Histone acetyltransferase KAT8 in the sample or standard competes with a fixed amount of biotin-labeled Histone acetyltransferase KAT8 for sites on a pre-coated Monoclonal antibody specific to Histone acetyltransferase KAT8. Excess conjugate and unbound sample or standard are washed from the plate. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Histone acetyltransferase KAT8 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

MYST1: Histone acetyltransferase which may be involved in transcriptional activation. May influence the function of ATM. As part of the MSL complex it is involved in acetylation of nucleosomal histone H4 producing specifically H4K16ac. As part of the NSL complex it may be involved in acetylation of nucleosomal histone H4 on several lysine residues. That activity is less specific than the one of the MSL complex. Belongs to the MYST (SAS/MOZ) family. 2 isoforms of the human protein are produced by alternative splicing.
