d cells. Gremlin/Drm belongs to the DAN family of secreted glycoproteins that are BMP antagonists. Other members of the family include: Cerberus, Dante, PRDC, Caronte and DAN. DAN family members share a cysteine-rich domain that is structurally related to the cysteine-knot motif found in TGFbeta superfamily ligands. In vitro, Gremlin/Drm binds BMP4 and BMP2 indicating that it might interfere with BMP signaling. Gremlin/Drm acts as a BMP2/ 4 antagonist in a variety of tissues and developmental processes including: Xenopus animal cap explants, chick limb bud outgrowth and chondrogenesis, murine lung branching morphogenesis, and osteogenic differentiation of mouse myoblasts and bone marrow stromal cells. In addition, expression of Gremlin/Drm has been shown to be down-regulated in a wide range of human cancer cell lines. Mouse, human, chick and Xenopus homologs of Gremlin share over 80% amino acid identity. It is likely that various DAN family members and other BMP antagonists including Noggin, Chordin, Follistatin and TSG can selectively antagonize the activities of different subsets of TGFbeta superfamily ligands.

Function: Cytokine that may play an important role during carcinogenesis and metanephric kidney organogenesis, as BMP a antagonist required for early limb outgrowth and patterning in maintaining the FGF4-SHH feedback loop. Down-regulates the BMP4 signaling in a dose-dependent manner. Acts as inhibitor of monocyte chemotaxis. Ref.4 Ref.5 Ref.6

Subunit structure: Interacts with SLIT1 and SLIT2 in a glycosylation-dependent manner

Tissue specificity: Highly expressed in spleen and to a lesser extent in lung, skeletal muscle and kidney. Expressed only in non-transformed cells or primary fibroblasts in culture but not in established transformed or tumor derived cell lines. Broadly expressed in limb bud mesenchyme but restricted to the distal limb bud mesenchyme and concentrated posteriorly. Expressed in ovary especially in granulosa cells of follicles of type 4. Induction: Up-regulated by GDF9 dose-dependent manner and BMP4 in granulosa cells. Highly regulated during folliculogenesis. Ref.5
