Solubility
It is recommended to reconstitute the lyophilized Macrophage Inflammatory Protein-1a in sterile 18M Omega -cm H2O not less than 100 ug/ml, which can then be further diluted to other aqueous solutions.
Biological Activity
The Activity is calculated by the ability to chemoattract of rat peritoneal macrophages using a conc. of 60 ng/ml corresponding to a Specific Activity of 16,667IU/mg.
Preparation and Storage
Lyophilized MIP-1a although stable at room temperature for 3 weeks, should be stored desiccated below -18 degree C. Upon reconstitution CCL3 should be stored at 4 degree C between 2-7 days and for future use below -18 degree C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Please prevent freeze-thaw cycles.
Related Product Information for rMIP 1a active protein
Description: Macrophage Inflammatory Protein-1 alpha Rat Recombinant produced in E Coli is a single, non-glycosylated, polypeptide chain containing 69 amino acids and having a molecular mass of 7853 Dalton. The MIP-1a is purified by proprietary chromatographic techniques.

Introduction: Macrophage Inflammatory Proteins (MIP) belong to the family of chemotactic cytokines known as chemokines. In humans, there are two major forms, MIP-1a and MIP-1b that are now officially named CCL3 and CCL4 respectively. Both are major factors produced by macrophages after they are stimulated with bacterial endotoxins. They activate human granulocytes (neutrophils, eosinophils and basophils) which can lead to acute neutrophilic inflammation. They also induce the synthesis and release of other pro-inflammatory cytokines such as interleukin 1 (IL-1), IL-6 and TNF-a from fibroblasts and macrophages. The genes for CCL3 and CCL4 are both located on human chromosome 17.

NCBI Description

mediates monocyte and neutrophil chemotaxis; may play a role in the pathogenesis of acute lung injury [RGD, Feb 2006]

Uniprot Description

Monokine with inflammatory and chemokinetic properties. Has chemotactic activity for monocytes, neutrophils, eosinophils, basophils, and lymphocytes. Required for lung TNF-alpha production, neutrophil recruitment and subsequent lung injury and may function as an autocrine mediator for the macrophage production of TNF-alpha which in turn up-regulates vascular adhesion molecules required for neutrophil influx. This protein binds heparin.

Product Notes

The rMIP 1a ccl3 (Catalog #AAA142050) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first five N-terminal amino acids was determined and was found to be, Ala-Pro-Ty r-Gly-Ala. It is sometimes possible for the material contained within the vial of "Macrophage Inflammatory protein-1 alpha, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
