Host Note
Optimized DNA sequence encoding Human MIP-3 alpha (CCL20) mature chain was expressed in Escherichia Coli.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note
Human CCL20, generated by the proteolytic removal of the signal peptide
and propeptide and has a calculated molecular mass of approximately 8 kDa.

Recombinant CCL-20 is a disulfide-linked monomeric protein consisting of 71 amino acid residue subunits and migrates as an approximately 8 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.
Related Product Information for MIP-3A recombinant protein
Macrophage inflammatory protein (MIP) -3/CCL20, also known as liver and activation-regulated chemokine (LARC) or Exodus, is a member of the CC chemokine subfamily initially noted to be expressed in human liver, lung, appendix, and tonsillar crypts. MIP-3 is selectively chemotactic for CD34+ bone marrow cell-derived immature DCs and CD45RO+ memory T cells that express the cognate receptor CCR6. MIP-3 produced at sites of inflammation may chemoattract CCR6-expressing immature DCs to the subepithelial region of mucosal surfaces.

NCBI Description

This antimicrobial gene belongs to the subfamily of small cytokine CC genes. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The protein encoded by this gene displays chemotactic activity for lymphocytes and can repress proliferation of myeloid progenitors. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2014]

Uniprot Description

CCL20: Chemotactic factor that attracts lymphocytes and, slightly, neutrophils, but not monocytes. Inhibits proliferation of myeloid progenitors in colony formation assays. May be involved in formation and function of the mucosal lymphoid tissues by attracting lymphocytes and dendritic cells towards epithelial cells. C-terminal processed forms have been shown to be equally chemotactically active for leukocytes. Possesses antibacterial activity E.coli ATCC 25922 and S.aureus ATCC 29213. Belongs to the intercrine beta (chemokine CC) family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Secreted, signal peptide; Secreted; Motility/polarity/chemotaxis; Chemokine

Chromosomal Location of Human Ortholog: 2q36.3

Cellular Component: extracellular space; extracellular region

Molecular Function: protein binding; chemokine activity

Biological Process: cell-cell signaling; wound healing; defense response to bacterium; immune response; inflammatory response; chemotaxis; positive regulation of interleukin-1 alpha biosynthetic process; signal transduction; positive regulation of nitric-oxide synthase biosynthetic process

Product Notes

The MIP-3A ccl20 (Catalog #AAA553195) is a Recombinant Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: MCCTKSLLLA ALMSVLLLHL CGESEAASNF DCCLGYTDRI LHPKFIVGFT RQLANEGCDI NAIIFHTKKK LSVCANPKQT WVKYIVRLLS KKVKNM. It is sometimes possible for the material contained within the vial of "MIP-3 alpha (CCL20), Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
