Mouse Mediator Of DNA Damage Checkpoint Protein 1 (MDC1) ELISA Kit | MDC1 elisa kit
Mouse Mediator Of DNA Damage Checkpoint Protein 1 (MDC1) ELISA Kit
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Background: The protein encoded by Mdc1 contains an N-terminal forkhead domain, two BRCA1 C-terminal (BRCT) motifs and a central domain with 7 divergent copies of an approximately 41-amino acid sequence. The encoded protein is required to activate the intra-S phase and G2/M phase cell cycle checkpoints in response to DNA damage. This nuclear protein interacts with phosphorylated histone H2AX near sites of DNA double-strand breaks through its BRCT motifs, and facilitates recruitment of the ATM kinase and meiotic recombination 11 protein complex to DNA damage foci. Mice with mutations in Mdc1 exhibit growth retardation, male infertility, immune defects, chromosome instability, DNA repair defects, and radiation sensitivity.

Uniprot Description
MDC1: Required for checkpoint mediated cell cycle arrest in response to DNA damage within both the S phase and G2/M phases of the cell cycle. May serve as a scaffold for the recruitment of DNA repair and signal transduction proteins to discrete foci of DNA damage marked by 'Ser-139' phosphorylation of histone H2AFX. Also required for downstream events subsequent to the recruitment of these proteins. These include phosphorylation and activation of the ATM, CHEK1 and CHEK2 kinases, and stabilization of TP53 and apoptosis. ATM and CHEK2 may also be activated independently by a parallel pathway mediated by TP53BP1. 4 isoforms of the human protein are produced by alternative splicing.
Protein type: Cell cycle regulation
Cellular Component: nucleoplasm; focal adhesion; chromosome; nucleus
Molecular Function: protein C-terminus binding

Product Notes
The Mouse MDC1 mdc1 (Catalog #AAA9717698) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9717698 ELISA Kit recognizes Mouse MDC1. It is sometimes possible for the material contained within the vial of "Mediator Of DNA Damage Checkpoint Protein 1 (MDC1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
