Intended Uses: This MDC1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken MDC1. This ELISA kit for research use only!

Principle of the Assay||MDC1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-MDC1 antibody and an MDC1-HRP conjugate. The assay sample and buffer are incubated together with MDC1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MDC1 concentration since MDC1 from samples and MDC1-HRP conjugate compete for the anti-MDC1 antibody binding site. Since the number of sites is limited, as more sites are occupied by MDC1 from the sample, fewer sites are left to bind MDC1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MDC1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene contains an N-terminal forkhead domain, two BRCA1 C-terminal (BRCT) motifs and a central domain with 13 repetitions of an approximately 41-amino acid sequence. The encoded protein is required to activate the intra-S phase and G2/M phase cell cycle checkpoints in response to DNA damage. This nuclear protein interacts with phosphorylated histone H2AX near sites of DNA double-strand breaks through its BRCT motifs, and facilitates recruitment of the ATM kinase and meiotic recombination 11 protein complex to DNA damage foci. Uniprot Description

MDC1: Required for checkpoint mediated cell cycle arrest in response to DNA damage within both the S phase and G2/M phases of the cell cycle. May serve as a scaffold for the recruitment of DNA repair and signal transduction proteins to discrete foci of DNA damage marked by 'Ser-139' phosphorylation of histone H2AFX. Also required for downstream events subsequent to the recruitment of these proteins. These include phosphorylation and activation of the ATM, CHEK1 and CHEK2 kinases, and stabilization of TP53 and apoptosis. ATM and CHEK2 may also be activated independently by a parallel pathway mediated by TP53BP1. 4 isoforms of the human protein are produced by alternative splicing.
