Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- MPO antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-MPO antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the MPO amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of MPO can be calculated.

Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of neutrophils. [provided by RefSeq, Nov 2014]

Homodimer; disulfide-linked. Each monomer consists of a light and a heavy chain. Belongs to the peroxidase family.
