Intended Uses: This MPO ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey MPO. This ELISA kit for research use only!

Principle of the Assay: MPO ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MPO antibody and an MPO-HRP conjugate. The assay sample and buffer are incubated together with MPO-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MPO concentration since MPO from samples and MPO-HRP conjugate compete for the anti-MPO antibody binding site. Since the number of sites is limited, as more sites are occupied by MPO from the sample, fewer sites are left to bind MPO-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MPO concentration in each sample is interpolated from this standard curve.

NCBI Description

Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of netrophils. [provided by RefSeq, Jul 2008]

MPO: Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity. Homodimer; disulfide-linked. Each monomer consists of a light and a heavy chain. Belongs to the peroxidase family. XPO subfamily. 3 isoforms of the human protein are produced by alternative splicing.
