Intended Uses: This sandwich kit is for the accurate quantitative detection of Porcine Myeloperoxidase (also known as MPO) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Porcine MPO antibody. MPO present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Porcine MPO Antibody is added and binds to MPO in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated MPO antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Porcine MPO. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of neutrophils. [provided by RefSeq, Nov 2014]

Uniprot Description

Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity.
