Related Product Information for MPO elisa kit
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-MPO polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-MPO polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the MPO amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of MPO can be calculated.!!Background/Introduction: Myeloperoxidase (MPO) is a peroxidase enzyme that in humans is encoded by the MPO gene which contains 12 exons and spans about 13 kb. Myeloperoxidase is most abundantly expressed in neutrophil granulocytes. It is a lysosomal protein stored in azurophilic granules of the neutrophil. The 150-kDa MPO protein is a dimer consisting of two 15-kDa light chains and two variable-weight glycosylated heavy chains bound to a prosthetic heme group. MPO produces hypochlorous acid (HOCl) from hydrogen peroxide (H2O2) and chloride anion (Cl-) during the neutrophil's respiratory burst. It requires heme as a cofactor. Furthermore, it oxidizes tyrosine to tyrosyl radical using hydrogen peroxide as an oxidizing agent.

NCBI Description

Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of netrophils. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity.

Catalytic activity: Cl- + H2O2 + H+ = HClO + H2O.Cl- + H2O2 = HOCl + 2 H2O.

Cofactor: Binds 1 calcium ion per monomer.Binds 1 heme B (iron-protoporphyrin IX) group covalently per monomer.

Subunit structure: Homodimer; disulfide-linked. Each monomer consists of a light and a heavy chain. Ref.13 Ref.21

Subcellular location: Lysosome.

Involvement in disease: Myeloperoxidase deficiency (MPOD) [MIM:254600]: A disorder characterized by decreased myeloperoxidase activity in neutrophils and monocytes that results in disseminated candidiasis.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.24 Ref.25 Ref.26 Ref.27 Ref.28

Sequence similarities: Belongs to the peroxidase family. XPO subfamily.

Product Notes

The Human MPO mpo (Catalog #AAA355337) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA355337 ELISA Kit recognizes Human MPO. AAA Biotech's Myeloperoxidase / MPO can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA). For quantitative detection of MPO in human serum, body fluids, tissue lysates or cell culture supernatants. Researchers should empirically determine the suitability of the MPO mpo for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Myeloperoxidase / MPO, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
