Rabbit Caspase-14 Polyclonal Antibody | anti-Casp14 antibody
Polyclonal Antibody to Caspase-14
IHC (paraffin): 1: 1000-1: 5000
IP: 1: 50-1: 200
For long-term storage, store at -20 degree C.
Avoid repeated freeze and thaw cycles.
Western Blot (WB)
(Fig: 1 Western blot analysis of Caspase-14. Tissue lysates (50 ug/lane) and recombinant human Caspase-14 were (Hu C14, 15 ng) were Fig: 1 Western blotted with Caspase-14 antibody at 1: 2000. The antibody detected both the proform of caspase-14, and the large and small subunits of active/cleaved caspase-14.)
Immunohistochemistry (IHC)
(Fig: 2 Formalin-fixed paraffin-embedded sections from a human ovarian cancer tissue microarray stained for Caspase-14 expression using at 1: 2000. Low (A) and high (B) stage ovarian tumor tissue cores. High magnification from areas of the tissue cores (A1 and B1). Decreased Caspase-14 expression was seen in the high grade, compared to the low grade tumor. Hematoxylin-eosin counterstain.)
Immunohistochemistry (IHC)
(Fig: 3 Formalin-fixed paraffin-embedded tissue sections of human cervix stained for Caspase-14 expression using at 1: 2000. A. Normal cervix (squamous epithelium). B. CIN1 (low-grade squamous intraepithelial lesion, mild dysplasia). C. CIN2 (high-grade squamous intraepithelial lesion, moderate dysplasia. D. CIN3 (high-grade squamous intraepithelial lesion; severe dysplasia-carcinoma in situ. In normal cervi, caspase-14 staining was found most in the midzone layer, but was absent from the basal/parabasal cell layer where mitotically active cells are known to reside. This suggests induction of caspase-14 expression with differentiation. Caspase-14 expression declined progressively during malignant transformation as the histologic severity of the cervical atypia advanced from CIN1 to CIN3. Hematoxylin-eosin counterstain.)
NCBI and Uniprot Product Information
Uniprot Description
CASP14: Believed to be a non-apoptotic caspase which is involved in epidermal differentiation. Seems to play a role in keratinocyte differentiation and cornification. Probably regulates maturation of the epidermis by proteolytically processing filaggrin. Complex of unprocessed caspase-14 and processed 19 kDa (p19) and 10 kDa (p10) subunits. In undifferentiated keratinocytes under postconfluency growth conditions (in vitro). Expressed in keratinocytes of adult skin suprabasal layers (from spinous layers to the stratum granulosum and stratum corneum). Expressed in keratinocytes of hair shaft and sebaceous glands. In psoriatic skin only expressed at very low levels. Belongs to the peptidase C14A family.
Protein type: EC 3.4.22.-; Protease
Chromosomal Location of Human Ortholog: 10|10 C1
Cellular Component: cytoplasm; extracellular matrix; keratin filament; nucleus
Molecular Function: peptidase activity
Biological Process: apoptosis; proteolysis

Product Notes
The Casp14 casp14 (Catalog #AAA668571) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Polyclonal Antibody to Caspase-14 reacts with Dog, Gerbil, Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Caspase-14 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC) Formalin/Paraffin, Immunoprecipitation (IP). WB: 1: 1000-1: 2000 IHC (paraffin): 1: 1000-1: 5000 IP: 1: 50-1: 200. Researchers should empirically determine the suitability of the Casp14 casp14 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Caspase-14, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
