Application Notes
Suitable for use in Flow Cytometry, Immunocytochemistry, Immunofluorescence, Immunohistochemistr (paraffin sections) and Western Blot.
Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4 degree C with gentle shaking, overnight.
Immunohistochemistry (Paraffin): 1:200. Requires antigen retrieval using ctirate method.
Immunocytochemistry/Immunofluorescence: 1:50
Flow Cytometry: 1:50

Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

P38 is the newest member of the MAP kinase family. It is a reactivating kinase. It is activated in response to inflammatory cytokines, endotoxins, and osmotic stress. p38 MAP kinase (MAPK), also called RK (3) or CSBP (2), is the mammalian homologue of the yeast HOG kinase and participates in a signaling cascade controlling cellular responses to cytokines and stress (1-4). Four isoforms of p38 MAP kinase, alpha, beta, gamma and delta, have been identified so far. Like the SAPK/JNK pathway, p38 MAP kinase is activated by a variety of cellular stresses including osmotic shock, inflammatory cytokines, lipopolysaccharides (LPS), UV light and growth factors (1-5). MKK3 and SEK activate p38 MAP kinase by phosphorylation at threonine 180 and tyrosine 182. Activated p38 MAP kinase has been shown to phosphorylate and activate MAPKAP kinase 2 (3) and to phosphorylate the transcription factors ATF-2 (5), Max (6) and MEF2 (5-8).
