Principle of the Assay: TXA2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TXA2 antibody and an TXA2-HRP conjugate. The assay sample and buffer are incubated together with TXA2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TXA2 concentration since TXA2 from samples and TXA2-HRP conjugate compete for the anti-TXA2 antibody binding site. Since the number of sites is limited, as more sites are occupied by TXA2 from the sample, fewer sites are left to bind TXA2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TXA2 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the G protein-coupled receptor family. The protein interacts with thromboxane A2 to induce platelet aggregation and regulate hemostasis. A mutation in this gene results in a bleeding disorder. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2009]

TXA2-R: Receptor for thromboxane A2 (TXA2), a potent stimulator of platelet aggregation. The activity of this receptor is mediated by a G-protein that activates a phosphatidylinositol-calcium second messenger system. In the kidney, the binding of TXA2 to glomerular TP receptors causes intense vasoconstriction. Activates phospholipase C. Isoform 1 activates adenylyl cyclase. Isoform 2 inhibits adenylyl cyclase. Defects in TBXA2R are the cause of susceptibility to bleeding disorder platelet-type 13 (BDPLT13). BDPLT13 is a disorder characterized by reduced platelet aggregation and a tendency to mild mucocutaneous bleeding. Belongs to the G-protein coupled receptor 1 family. 2 isoforms of the human protein are produced by alternative splicing.
