Principle of the Assay: This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with TXA2. During the reaction, TXA2 in the sample or standard competes with a fixed amount of TXA2 on the solid phase supporter for sites on the Biotinylated Detection Ab specific to TXA2. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) are added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of TXA2 in the samples is then determined by comparing the OD of the samples to the standard curve.

This gene encodes a member of the G protein-coupled receptor family. The protein interacts with thromboxane A2 to induce platelet aggregation and regulate hemostasis. A mutation in this gene results in a bleeding disorder. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2009]

TXA2-R: Receptor for thromboxane A2 (TXA2), a potent stimulator of platelet aggregation. The activity of this receptor is mediated by a G-protein that activates a phosphatidylinositol-calcium second messenger system. In the kidney, the binding of TXA2 to glomerular TP receptors causes intense vasoconstriction. Activates phospholipase C. Isoform 1 activates adenylyl cyclase. Isoform 2 inhibits adenylyl cyclase. Defects in TBXA2R are the cause of susceptibility to bleeding disorder platelet-type 13 (BDPLT13). BDPLT13 is a disorder characterized by reduced platelet aggregation and a tendency to mild mucocutaneous bleeding. Belongs to the G-protein coupled receptor 1 family. 2 isoforms of the human protein are produced by alternative splicing.
