The binding activity of TNFRSF11A with TRAF5.RANK tumor necrosis factor receptor superfamily member 11A protein (TNFRSF11A) also known as receptor Activator of Nuclear Factor kappa B (RANK) or TRANCE Receptoris a member of the tumor necrosis factor receptor (TNFR) molecular sub-family. TNFRSF11A is the receptor for RANK-Ligand (RANKL) and part of the RANK/RANKL/OPG signaling pathway that regulates osteoclast differentiation and activation. It is associated with bone remodeling and repair, immune cell function, lymph node development, thermal regulation, and mammary gland development. Besides, TNF Receptor Associated Factor 5 (TRAF5) has been identified as an interactor of TNFRSF11A, thus a binding ELISA assay was conducted to detect the interaction of recombinant human TNFRSF11A and recombinant human TRAF5. Briefly, TNFRSF11A were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to TRAF5-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-TNFRSF11A pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of TNFRSF11A and TRAF5 was shown in Figure 1, and this effect was in a dose dependent manner.)

Immunohistochemistry (IHC)
(Immunohistochemical analysis of paraffin-embedded human transitional cell carcinoma of bladder, using NF-kB p65 Antibody(MBS179238)RELA was detected in paraffin-embedded tissue section. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1ug/ml rabbit anti-RELA Antibody (MBS179238)overnight at 4 degree C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37 degree C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen. )

(Western blot analysis of Phospho-NF-Kappa B p65 (S529) expression in HeLa cell lysates treated with Calyculin A and TNF-alpha (MBS1750229).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-RELA monoclonal antibody overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for RELA )
