Principle of the Assay: Rat Myogenin ELISA Kit employs a two-site sandwich ELISA to quantitate Myog in samples. An antibody specific for Myog has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Myog present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Rat Myog detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of Myog bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background/Introduction: Myog acts to regulate myoblast terminal differentiation, myofibre maturation and size. Genetic studies revealed that among MRFs Myog is essential for viability. Extracellular factors determine the need for Myog function. Congruently, Myog myoblasts efficiently contribute to multinucleated fibres in genetic mosaic experiments. Furthermore, depletion of Myog after birth reduces myofibre size and affects overall body homeostasis. However, given that other MRFs can and do bind the same DNA motifs as Myog, the precise role(s) of Myog remain ill-defined.
