Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Intended Uses: The kit is a competitive inhibition enzyme immunoassay technique for the in vitro quantitative measurement of cholecystokinin in rat serum, plasma and other biological fluids.

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific to cholecystokinin has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled cholecystokinin and unlabeled cholecystokinin (Standards or samples) with the pre-coated antibody specific to cholecystokinin. After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of cholecystokinin in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of cholecystokinin in the sample.

Uniprot Description

Function: This peptide hormone induces gall bladder contraction and the release of pancreatic enzymes in the gut. Its function in the brain is not clear. Binding to CCK-A receptors stimulates amylase release from the pancreas, binding to CCK-B receptors stimulates gastric acid secretion.

Subunit structure: Binds to CCK-A receptors in the pancreas and CCK-B receptors in the brain

Tissue specificity: The shortest form (CCK8) is predominantly found in the brain, whereas the larger ones are found in the intestine.

Post-translational modification: The precursor is cleaved by proteases to produce a number of active cholecystokinins.Sulfation of Tyr-97 is essential for receptor activation.

Sequence similarities: Belongs to the gastrin/cholecystokinin family.
