Assay Procedure Step 1
Aliquot 0.1ml per well of the 10000pg/ml, 5000pg/ml, 2500pg/ml, 1250pg/ml, 625pg/ml, 312pg/ml, 156pg/ml mouse EPCR standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffer into the control well (Zero well). Add 0.1ml of each properly diluted sample of mouse cell culture supernates, serum or plasma(heparin, EDTA) to each empty well. It is recommended that each mouse EPCR standard solution and each sample be measured in duplicate.
Preparation and Storage
Store at 4 degree C for 6 months, at -20 degree C for 12 months. Avoid multiple freeze-thaw cycles.

For quantitative detection of activated mouse EPCR in cell culture supernates, cell lysates, tissue homogenates, serum and plasma (heparin, EDTA).
Background: Endothelial protein C receptor (EPCR), also known as CCCA or PROCR, is a protein that in humans is encoded by the PROCR gene. PRPCR gene is mapped to 20q11.2 by radiation hybrid analysis and FISH. The protein encoded by this gene is a receptor for activated protein C, a serine protease activated by and involved in the blood coagulation pathway. The encoded protein is an N-glycosylated type I membrane protein that enhances the activation of protein C. Mutations in this gene have been associated with venous thromboembolism and myocardial infarction, as well as with late fetal loss during pregnancy.
Principle: Mouse EPCR ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from Rat specific for EPCR has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: L18-S214) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for EPCR is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substRate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Mouse EPCR amount of sample captured in plate.

Uniprot Description

EPCR: a single-pass type I membrane protein. Binds activated protein C (APC), a serine protease that regulates thrombin (IIa) production. Plays a pivotal role in preventing blood coagulation through binding of activated protein C. Expressed strongly in the endothelial cells of arteries and veins in heart and lung, less intensely in capillaries in the lung and skin, and not at all in the endothelium of small vessels of the liver and kidney. The binding of APC to EPCR and PAR-1 increases invasion and chemotaxis.
