Application Notes
Anti-TIMP-1 works well for Western Blotting applications, and when used against the reduced protein identifies a band at 29 Kd. RP3T1 recognizes both reduced and non-reduced protein, and has applications for immunoprecipitation, immunohistochemistry, and ELISA applications. TIMP-1 is produced in low (pg/ml) levels by most cell types. Treatment of cells with the phorbol ester TPA stimulates production of TIMP-1 in some cell types, but the low protein levels produced often require concentration of cell culture media to visualize the bands by Western Blotting. A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples. RP3T1 recognizes a wider species range of TIMP-1 than does RP1T1.

Preparation and Storage
This product is stable for several weeks at 4 degree C as an undiluted liquid. Dilute only prior to immediate use.
For extended storage, aliquot contents and freeze at-20 degree C or below. Avoid cycles of freezing and thawing. Expiration date is one (1) year from date of receipt.

Description: The antibody is a Rabbit antibody to the Tissue Inhibitor of Metalloproteinases, TIMP-1, made with a synthetic peptide based on the carboxyterminal region of the Human sequence. The antibody binds to TIMP-1, but does not cross react with the other TIMP family members (TIMP-2, TIMP-3, TIMP-4). The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of Sodium Azide as preservative and 50% glycerol as cryoprotectant.

Relevance: Tissue inhibitor of metalloproteinases 1 (TIMP1) and Tissue inhibitor of metalloproteinases 2 (TIMP 2) have similar properties, specifically in inhibiting enzymes of matrix metalloproteinase family, and are thought to be of great importance in the maintenance of connective tissue integrity. TIMP1 forms a complex of 1:1 stoichiometry with activated interstitial collagenases, activated stromelysin, active form of 72kDa Type IV collagenase (also known as MMP2 or gelatinase A), and latent and active forms 92kDa Type IV collagenase (also known as MMP0 or gelatinase B). TIMPs inhibit the proteolytic invasiveness of tumour cells and normal placental trophoblast cells. TIMP1 is produced in low (pg/ml) levels by most cell types. Treatment of cells with the phorbol ester TPA stimulates production of TIMP1 in some cell types, but the low protein levels produced often require concentration of cell culture media to visualize the bands by Western Blotting.

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TIMP-1 (Tissue Inhibitor of Metalloproteinases), Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
