Application Notes
The antibody works well for Western Blotting applications, and when used against the reduced protein identifies bands at 65 Kd and 63 Kd (the pro-form and active form), as well as activation/breakdown products_recover. The antibody can recognize native or reduced MT1MMP, and has application for immunoprecipitation, immunohistochemistry, cell sorting and ELISA. MT1MMP has been reported to be elevated in several tumor cell lines, and is constituitively produced by some normal cell lines. Treatment of cells with Concanavolin-A or the phorbol ester TPA stimulates production of MT1MMP in some cell types, and the enzyme can be recovered in cell lysates. Shed forms of MT1MMP have also been reported. A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples.

Description: The antibody is a Rabbit antibody to the membrane-bound Matrix Metalloproteinase-1 (MMP-14), made with a synthetic peptide based on the Human sequence. The antibody binds to the hinge region of MT1MMP. The antibody binds to MT1MMP, but does not cross react with the other MMP family members (MMP1, MMP2, MMP3, MMP-9). The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of Sodium Azide as preservative, and 50% glycerol as cryoprotectant.

Relevance: The matrix metalloproteinases (MMPs) are a family of at least eighteen secreted and membrane bound zincendopeptidases. Collectively, these enzymes can degrade all the components of the extracellular matrix, including fibrillar and non fibrillar collagens, fibronectin, laminin and basement membrane glycoproteins. In general, a signal peptide, a propeptide, and a catalytic domain containing the highly conserved zinc binding site characterizes the structure of the MMPs. In addition, fibronectin like repeats, a hinge region, and a C terminal hemopexin like domain allow categorization of MMPs into the collagenase, gelatinase, stomelysin and membrane-type MMP subfamilies. All MMPs are synthesized as proenzymes, and most of them are secreted from the cells as proenzymes. Thus, the activation of these proenzymes is a critical step that leads to extracellular matrix breakdown. MMPs are considered to play an important role in wound healing, apoptosis, bone elongation, embryo development, uterine involution, angiogenesis and tissue remodeling, and in diseases such as multiple sclerosis, Alzheimer's, malignant gliomas, lupus, arthritis, periodontis, glumerulonephritis, atherosclerosis, tissue ulceration, and in cancer cell invasion and metastasis. MMP14 may be an activator of pro gelatinase A and is expressed in fibroblast cells during both wound healing and human cancer progression. MMP14 is expressed in very low levels and may require stimulation with concanavolin A or the phorbol ester TPA to stimulate production of MMP14.

Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. However, the protein encoded by this gene is a member of the membrane-type MMP (MT-MMP) subfamily; each member of this subfamily contains a potential transmembrane domain suggesting that these proteins are expressed at the cell surface rather than secreted.
