Immunohistochemistry
Synonyms
SPHK1; Polyclonal Antibody; RABBIT ANTI HUMAN SPHK1 (N-TERMINAL); anti-SPHK1 antibody
Ordering
For Research Use Only!
Host
Rabbit
Clonality
Polyclonal
Isotype
IgG
Specificity
This item detects an epitope within the N-terminal region human SphK1, also known sphingosine kinase 1. SphK1 catalyzes the phosphorylation of sphingosine 1-phosphate; an important lipid mediator with multiple functions. In light of this, SphK1 has been implicated in regulation of Ca2+ homoeostasis, inflammation, immunity and cancer growth. However, its central function is thought to be in the development of the cardiovascular and central nervous system. SphK1 has a molecular weight of 42kD.

Applicable Applications for anti-SPHK1 antibody
Immunohistology Paraffin*
Application Notes
Immunohistology - Paraffin: Minimum Dilution: 5; Maximum Dilution: 15ug/ml; Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose.
Perservative Stabilisers
0.1% Sodium Azide (NaN3)
Immunogen
Synthetic peptide: N-terminal domain of human SphK1, conjugated to KLH.
Histology Positive Control Tissue
Antiserum Preparation
Antisera to SphK1 were raised by repeated immunisations of rabbit with highly purified antigen. Purified IgG prepared by affinity chromatography.
Target Species
Human
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 18 months from date of despatch.

NCBI Description

The protein encoded by this gene catalyzes the phosphorylation of sphingosine to form sphingosine-1-phosphate (S1P), a lipid mediator with both intra- and extracellular functions. Intracellularly, S1P regulates proliferation and survival, and extracellularly, it is a ligand for cell surface G protein-coupled receptors. This protein, and its product S1P, play a key role in TNF-alpha signaling and the NF-kappa-B activation pathway important in inflammatory, antiapoptotic, and immune processes. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2011]

Uniprot Description

SPHK1: Catalyzes the phosphorylation of sphingosine to form sphingosine 1-phosphate (SPP), a lipid mediator with both intra- and extracellular functions. Also acts on D-erythro-sphingosine and to a lesser extent sphinganine, but not other lipids, such as D,L-threo-dihydrosphingosine, N,N-dimethylsphingosine, diacylglycerol, ceramide, or phosphatidylinositol. Interacts with ACY1. Binds to calmodulin. Interacts with SPHKAP. Widely expressed with highest levels in adult liver, kidney, heart and skeletal muscle. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The SPHK1 sphk1 (Catalog #AAA221451) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's SPHK1 can be used in a range of immunoassay formats including, but not limited to, Immunohistology Paraffin*. Immunohistology - Paraffin: Minimum Dilution: 5; Maximum Dilution: 15ug/ml; Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose. Researchers should empirically determine the suitability of the SPHK1 sphk1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SPHK1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
